Mechanism of Action: The polyene antiobiotics (amphotericin, nystatin) bind to sterols on fungal cell membranes to deform the cells. the imidazoles (clotrimazole, ketoconazole, miconazole, isoconazole, tioconazole) cause lethal accumulation of hydrogen peroxide in fungal cell

Precautions: Hypersensitivity. liver disease. co administration with terfenadine or cisapride.

Indications/Uses: Candida infections 50-200mg daily, with or without food. crytococcal meningitis 200-400mg daily.

Interactions: Increases effect of theophylline, anticoagulants, cyclosporin, oral hypoglycaemics, terfenadine. reduced effect: rifampicin.
